Characterization of functional domains of starfish and mollusk mannose 6-phosphate receptor protein (MPR300)


Current advances

Bhamidimarri/Siva Kumar will work on the analysis of functional domains of the Mannose 6-phosphate/IGF-II receptor from the invertebrate starfish and Unio. Although the receptors have been purified and preliminary biochemical studies done by us for the first time, there is no information on their structural similarities to the well characterized vertebrate receptors. This is essential to be obtained to support the hypothesis on the evolutionary conservation of the lysosomal biogenesis pathway in invertebrates, as we have preliminary results indicating that the lysosomal enzyme α-fucosidase from starfish is structurally related to the human enzyme. In addition, she will also purify α-mannosidase from Unio and analyze its partial sequence and N-glycan analysis (col. Mormann). The results obtained will establish the evolution of the lysosomal enzyme sorting receptors in the invertebrates.
