Interplay of Syndecan-1 and heparanase in colon cancer pathogenesis

K. Kumar /Götte investigated the functional interplay of the HS degrading enzyme heparanase, and its substrate Syndecan-1 in colon cancer using in vitro siRNA and mutant/wild-type plasmid over-expression approaches. We report for the first time the involvement of Sdc1 in regulating Heparanase expression at the promoter level. Interestingly, we were able to show the impact of Sdc1 and HPSE on a cancer stem cell phenotype via a Wnt signaling-dependent mechanism. Further characterization of biochemical growth factor-, morphogen- and integrin-Sdc1 interactions are being explored in cooperation with the groups of Mormann and Swamy.
