Structural elucidation of proteoglycans


Glycans and proteoglycans exert manifold cellular effects e.g. the modulation of skin-cell activity, regulation of immune cell activity, cell-cell signalling etc. Within the ongoing project proteoglycans from Jatropha curcas are analysed structurally for a better understanding of functional activity towards human keratinocytes.
Beside optimisation of extraction procedure fractionation of the complex proteoglycan mixtures will lead to highly purified polymers. Detailed structural analysis of the carbohydrate part has to be performed by means of HPAEC-PAD, GLC-MS, NMR, MS etc. as well as analysis of the protein moiety. Additionally lectins from J. curcas will be analysed concerning specificity and molecular features.


Current advances


In collaboration with Mormann/König/Swamy developed a validated bunch of analytical methods for investigating the fine structures and glycosylation sides of arabinogalactan proteins (AGP).

The project (dissertation finalized, manuscripts submitted) provides validated tools for fine-structure analytical characterization of AGPs and understanding of the role of AGP clusters in embryo tissues.
