N-Glycoproteomics of the whey fraction of human milk

Current advances


We have started investigations on the structural variability of N-glycans derived from human milk glycoproteins. Currently, sophisticated protocols for a comprehensive analysis of the human milk glycoproteome are being developed. This challenging approach is aimed at the isolation of glycoproteins, followed by fractionation. Regarding specific prefractionation, differential lectin affinity chromatography will be employed, including a wealth of lectins with different specificities in order to achieve a prefractionation of the glycoproteome aimed at enrichment of glycoproteins prior to MS analyses. The collaboration with Prof. Siva Kumar at the University of Hyderabad is envisaged essentially for this part of the project, since this lab is highly expertised in the purification of glycoconjugates and the application of plenty of plant and invertebrate lectins for this purpose. Subsequent analyses of glycopeptides upon digestion with proteases of varying specificity will give rise to information on glycan structures and the corresponding glycosylation sites. In collaboration with Hensel and Müthing, we intend to explore the protective effects of milk glycoproteins and glycolipids against pathogenic bacteria and pathogen-released toxins.
